My Mutation Calculator can help you figure out things like this. Plug in 5
transmission events and 25 markers and use .002 for the mutation rate. The
probability of observing 2 mutations like this is about 2%.

Also, although we don't have enough data to be confident, it does appear that
your mutations occurred in markers with higher than average mutation rates,
DYS389-2 and DYS449.
