I have just recruited an uncle (my fathers brother) for testing, so this
could shed some light on the matter.

My Mutation Calculator can help you figure out things like this. Plug in
5
transmission events and 25 markers and use .002 for the mutation rate.
The
probability of observing 2 mutations like this is about 2%.
