Hi Kathy,
I don't think you or your family have anything to worry about. T4336C has not been observed in U5 or any subclade to date. It does, however appear in U6, especially the U6d clade. (cf. A Comprehensive hg U Mutation List and other sequences - http://tinyurl.com/yyzgaa under mtDNA).
