"Flintstone PO" stands for "Flintstone Post Office", which is a geographic/locational feature used by the U.S. Geologic Survey and others for map indices. In earlier times, before the days of home delivery, since the post office was usually found in the center of a town or village, and was (is) an official federal government office, map makers commonly used its location to indicate the center of a community, even though the post office may have been inside or part of another building, such as a general store. Any good road atlas will show Flintstone east of Cumberland on I-68.
(I agree with Connie, it's about 10 miles). The name may have originally been Flint Stone (two words) because of the high amount of flint and shale in the surrounding hills, but over time it became truncated. No relation to Fred, Wilma, or Pebbles...
